Thank you all for your valuable insights. I actually work on plants, but I have the similar issue preparing samples for 10X Visium Spatial Gene Expression. It always gives me cracking tissue slices when I use isopentane/liquid freezing. Only slow freezing (at -20C) gives me intact slices. I guess the isopentane/dry ice freezing might be worth a try for good quality RNA.
